An overall evaluation of the resistance (R) and pathogenesis-related (PR) superfamilies in soybean, as compared with Medicago and Arabidopsis Genet. Mol. Biol.
Wanderley-Nogueira,Ana C.; Belarmino,Luis C.; Soares-Cavalcanti,Nina da M.; Bezerra-Neto,João P.; Kido,Ederson A.; Pandolfi,Valesca; Abdelnoor,Ricardo V.; Binneck,Eliseu; Carazzole,Marcelo F.; Benko-Iseppon,Ana M..
Plants have the ability to recognize and respond to a multitude of pathogens, resulting in a massive reprogramming of the plant to activate defense responses including Resistance (R) and Pathogenesis-Related (PR) genes. Abiotic stresses can also activate PR genes and enhance pathogen resistance, representing valuable genes for breeding purposes. The present work offers an overview of soybean R and PR genes present in the GENOSOJA (Brazilian Soybean Genome Consortium) platform, regarding their structure, abundance, evolution and role in the plantpathogen metabolic pathway, as compared with Medicago and Arabidopsis. Searches revealed 3,065 R candidates (756 in Soybean, 1,142 in Medicago and 1,167 in Arabidopsis), and PR candidates matching to 1,261 sequences...

In silico identification of known osmotic stress responsive genes from Arabidopsis in soybean and Medicago Genet. Mol. Biol.
Soares-Cavalcanti,Nina M.; Belarmino,Luis C.; Kido,Ederson A.; Wanderley-Nogueira,Ana C.; Bezerra-Neto,João P.; Cavalcanti-Lira,Rafaela; Pandolfi,Valesca; Nepomuceno,Alexandre L.; Abdelnoor,Ricardo V.; Nascimento,Leandro C.; Benko-Iseppon,Ana M..
Plants experience various environmental stresses, but tolerance to these adverse conditions is a very complex phenomenon. The present research aimed to evaluate a set of genes involved in osmotic response, comparing soybean and medicago with the well-described Arabidopsis thaliana model plant. Based on 103 Arabidopsis proteins from 27 categories of osmotic stress response, comparative analyses against Genosoja and Medicago truncatula databases allowed the identification of 1,088 soybean and 1,210 Medicago sequences. The analysis showed a high number of sequences and high diversity, comprising genes from all categories in both organisms. Genes with unknown function were among the most representative, followed by transcription factors, ion transport...

In silico survey of resistance (R) genes in Eucalyptus transcriptome Genet. Mol. Biol.
Barbosa-da-Silva,Adriano; Wanderley-Nogueira,Ana C.; Silva,Raphaela R.M.; Berlarmino,Luiz C.; Soares-Cavalcanti,Nina M.; Benko-Iseppon,Ana M..
A major goal of plant genome research is to recognize genes responsible for important traits. Resistance genes are among the most important gene classes for plant breeding purposes being responsible for the specific immune response including pathogen recognition, and activation of plant defence mechanisms. These genes are quite abundant in higher plants, with 210 clusters found in Eucalyptus FOREST database presenting significant homology to known R-genes. All five gene classes of R-genes with their respective conserved domains are present and expressed in Eucalyptus. Most clusters identified (93) belong to the LRR-NBS-TIR (genes with three domains: Leucine-rich-repeat, Nucleotide-binding-site and Toll interleucine 1-receptor), followed by the...

Intra- and interspecific chromosome polymorphisms in cultivated Cichorium L. species (Asteraceae) Genet. Mol. Biol.
Bernardes,Ebenézer C.S.; Benko-Iseppon,Ana M.; Vasconcelos,Santelmo; Carvalho,Reginaldo; Brasileiro-Vidal,Ana C..
Endive (Cichorium endivia L.) and chicory (C. intybus L.) both have 2n = 18, but until now, there has been no detailed karyomorphological characterization. The present work evaluated five accessions of each species using FISH with rDNA probes and fluorochrome staining with CMA and DAPI. Both species presented distinct banding patterns after fluorochrome staining: while endive had proximal CMA++/DAPI- bands in the short arms of pairs 1, 2 and 3, chicory had proximal CMA-positive bands in chromosomes 1 and 3 and interstitial in the short arm of chromosome 8. Among endive accessions, FISH procedures revealed conserved position and number of 5S and 45S rDNA sites (two and three pairs, respectively), associated with the CMA-positive bands. Notwithstanding,...

Mining plant genome browsers as a means for efficient connection of physical, genetic and cytogenetic mapping: an example using soybean Genet. Mol. Biol.
Belarmino,Luis C.; Oliveira,Ana R. da S.; Brasileiro-Vida,Ana C.; Bortoleti,Kyria C. de A.; Bezerra-Neto,João Pacífico; Abdelnoor,Ricardo V.; Benko-Iseppon,Ana M..
Physical maps are important tools to uncover general chromosome structure as well as to compare different plant lineages and species, helping to elucidate genome structure, evolution and possibilities regarding synteny and colinearity. The increasing production of sequence data has opened an opportunity to link information from mapping studies to the underlying sequences. Genome browsers are invaluable platforms that provide access to these sequences, including tools for genome analysis, allowing the integration of multivariate information, and thus aiding to explain the emergence of complex genomes. The present work presents a tutorial regarding the use of genome browsers to develop targeted physical mapping, providing also a general overview and examples...

Overall picture of expressed Heat Shock Factors in Glycine max, Lotus japonicus and Medicago truncatula Genet. Mol. Biol.
Soares-Cavalcanti,Nina M.; Belarmino,Luís C; Kido,Ederson A.; Pandolfi,Valesca; Marcelino-Guimarães,Francismar C.; Rodrigues,Fabiana A.; Pereira,Gonçalo A.G.; Benko-Iseppon,Ana M..
Heat shock (HS) leads to the activation of molecular mechanisms, known as HS-response, that prevent damage and enhance survival under stress. Plants have a flexible and specialized network of Heat Shock Factors (HSFs), which are transcription factors that induce the expression of heat shock proteins. The present work aimed to identify and characterize the Glycine max HSF repertory in the Soybean Genome Project (GENOSOJA platform), comparing them with other legumes (Medicago truncatula and Lotus japonicus) in view of current knowledge of Arabidopsis thaliana. The HSF characterization in leguminous plants led to the identification of 25, 19 and 21 candidate ESTs in soybean, Lotus and Medicago, respectively. A search in the SuperSAGE libraries revealed 68...

Updating the list of chromosome numbers for Philodendron (Araceae) Acta Botanica
Vasconcelos,Emanuelle V.; Brasileiro-Vidal,Ana C.; Benko-Iseppon,Ana M.; Vasconcelos,Santelmo.
ABSTRACT Aiming for a better understanding of karyotype evolution within Philodendron, we report chromosome counts for 23 species of the genus, of which 19 are being reported for the first time, thus increasing to 84 ( ca. 17 % of the genus) the total number of species with available chromosome counts. The diploid numbers 2 n = 32 and 2 n = 34 were the most common, with 10 and 11 species, respectively, whereas only two species presented different chromosome numbers ( P. giganteum with 2 n = 30 and P. adamantinum with 2 n = 36). The results are discussed in the context of previous analyses of karyotypes of Philodendron spp., taking into account bidirectional dysploidy as the main mechanism of chromosome number evolution within the genus.
